Osteogenic differentiation of mesenchymal stem cells through modulation of Wnt signaling

Background: Mesenchymal stem cells have considerable potential for bone regeneration, although their osteogenic differentiation is regulated by multiple signaling pathways. This study investigated the effect of Wnt pathway modulation on osteogenic differentiation.Methods: Mesenchymal stem cells were cultured under osteogenic conditions with or without a Wnt signaling modulator. Cell proliferation, alkaline phosphatase activity, mineral deposition, and osteogenic marker expression were evaluated.Results: Wnt pathway modulation enhanced alkaline phosphatase activity and extracellular matrix mineralization, accompanied by increased expression of RUNX2 and osteocalcin.Conclusion: Modulation of Wnt signaling may promote osteogenic differentiation and represents a potential strategy for bone tissue engineering.
